I’d check the following:
If you’re running Vista (or the Win 7 RC, etc.), right-click the Vent shortcut, select Properties, then go to the Compatibility tab. Check the box for “Run this program…” and select Windows XP Service Pack 2. Click Apply, then OK. Try using Vent.
Make sure the key you use for PTT isn’t already mapped to something in WoW.
Disable the in-game voice chat system if no one in your party, etc. uses it, just in case there’s a conflict.
